Adamax vs. Semax: One Has a Structure, One Has Three

Adamax is sold as an upgraded Semax that lasts longer and reaches the brain more easily. It might be. But the suppliers selling it don't agree on what molecule it is, and one of them lists a formula with no adamantane in it at all.

These two come up together constantly in conversations about focus and mental clarity. Someone asks what people use, a few answers come back with Semax, and then somebody adds that Adamax is the better version. Stronger, longer lasting, less of it needed. It sounds like a straightforward upgrade.

It isn't that kind of comparison. Semax has a known structure, a published literature, and a regulatory history in one country. Adamax is described three different ways by three different sources, and at least one supplier's own paperwork lists a molecule that doesn't contain the ingredient it's named after.

What they have in common

Adamax is built on Semax, so the family resemblance is genuine. Semax is a short peptide, seven amino acids long, derived from a fragment of a hormone called adrenocorticotropic hormone. Its sequence is methionine, glutamic acid, histidine, phenylalanine, proline, glycine, proline. Adamax takes that same backbone and modifies it, which is why anything people say about how Adamax supposedly works is really a description of Semax with an asterisk attached.

Both get researched in the same territory, meaning attention, memory, and protection of brain tissue after injury, and both are sold as research compounds outside Russia.

Where Semax comes from

Semax was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. It's been a registered pharmaceutical in Russia for decades, prescribed as a nasal spray for conditions including stroke recovery, optic nerve disease, and cognitive decline. That approval matters less as a stamp of quality than as an explanation for why the research exists at all. A country that licensed the drug kept funding studies of it.

The mechanism people cite most often is brain-derived neurotrophic factor, a protein involved in keeping neurons alive and helping them form new connections. Research in rats found that a single dose of Semax raised that protein in the hippocampus by roughly 1.4 times, and a separate study found Semax binds specifically in the basal forebrain and raises it there too. That's the pathway underneath most claims about focus and clarity.

The limits are worth stating. Most of that work is in animals, most comes from a small number of Russian institutions, and independent replication outside that circle is thin. The human data is modest and mostly in stroke patients rather than healthy people looking for sharper attention. There's also a 2007 paper proposing Semax for attention deficit hyperactivity disorder that people sometimes cite as if it were a trial result. It ran in Medical Hypotheses, a journal that exists to publish ideas worth considering rather than experiments that were run, and nothing has followed up on it in the nineteen years since.

Where Adamax comes from

This is where most write-ups get it wrong. A number of sites say Adamax emerged from Russian nootropic peptide research, which describes Semax, not Adamax.

Adamax was created and named by Ceretropic, a Mexico-based research chemical company. Ceretropic was effectively the only reliable source for it, and the company shut down permanently in 2018. So the compound's originator has been gone for eight years, and what's sold under the name today is being made by suppliers who weren't involved in designing it.

The design idea itself is reasonable and worth crediting. Semax breaks down fast in the body, so the modification adds an acetyl group to slow enzyme degradation and an adamantane group, a rigid cage-shaped hydrocarbon, to make the molecule better at crossing into the brain. Adamantane isn't an exotic trick, it's the same scaffold used in amantadine and memantine. On paper this is a sensible thing to try.

The part that actually matters

When we went looking for Adamax's structure, we didn't find one answer. We found three, and they aren't small variations on each other.

The community and vendor consensus describes an acetylated Semax with an Ala-Gly extension and an adamantyl group at the C-terminus. Work that formula out and it comes to about 1117 daltons.

BOC Sciences, a catalog supplier that sells Adamax, lists it as C44H61N11O13S at 984.10 daltons, sequence Ac-MEHFPGPAG. We ran that formula ourselves. N-acetyl Semax extended by alanine and glycine, as a free acid, comes to exactly C44H61N11O13S at 984.11 daltons. That's their number to two decimal places, so their listing is internally consistent. It also means there's no adamantane in it, and their full chemical name and structural identifier string contain no adamantyl cage either. The compound named after adamantane is catalogued, by a company selling it, as a molecule that doesn't have any.

Wikipep, which presents itself as a peptide encyclopedia, gives a third structure with the adamantane on the opposite end of the molecule, and a formula of C49H73N11O12. That one can be ruled out on sight. It contains no sulfur, and the Semax backbone starts with methionine, which contains a sulfur atom. Any molecule built on Semax has to have sulfur in it. The entry labels the formula approximate, but a formula missing an element the structure requires isn't an approximation of the right answer.

That same entry lists a half-life of four to eight hours in a tidy table next to Semax's thirty minutes. It reads like measured data. It isn't. It's inferred from how adamantane behaves in other drugs, and the four sources cited are three Semax papers and a general review of adamantane chemistry. Not one of them is about Adamax.

What's actually been published on Adamax

Nothing. We searched PubMed and every result for the term is a machine learning optimizer that happens to share the name, showing up in papers about tumor imaging and skin lesion classifiers. Searching by structure returns nothing either. There's no animal study, no human study, no pharmacokinetic measurement, no toxicology.

That absence isn't damning on its own. Plenty of compounds people research seriously run ahead of their literature, and we'd have a very short website if we only covered things with full trial records. But it does mean every specific number attached to Adamax, the half-life, the potency multiplier, the dosing range, was reasoned out from Semax and from adamantane rather than measured. Those are predictions, and they may turn out to be right. They just haven't been checked.

Bottom line

If the question is which of these two you can actually know something about, it's Semax, and it isn't close. Its evidence base has real gaps, mostly single-region research with thin human data. But the structure is settled, the studies are readable, and you can check any claim against a source.

Adamax is a reasonable idea that never got tested. It might be exactly what people say it is. The problem isn't that it's unproven, it's that you can't currently establish what you'd be buying. When two suppliers describe the same product name with a 130 dalton difference between them, that's not a rounding disagreement, it's two different molecules sharing a label.

That also breaks price comparison, which is usually where we'd land. Per-milligram pricing only means something if a milligram is the same thing everywhere, and here it isn't. If you're looking at Adamax anyway, ask the vendor for a certificate of analysis with mass spectrometry data and check whether the measured mass matches what they claim to sell. That's a normal thing for a good supplier to have, and if they can't produce one, you've learned something worth knowing.
