Somebody Put Old Cells in Young Blood, and They Started Acting Young

The copper peptide in half the serums on the shelf began as a strange result in a 1973 liver experiment. What it turned out to be is stranger than anything on the label, and it explains why a scrape takes so much longer to close now than it used to.

In 1973 a biochemist named Loren Pickart ran an experiment that sounds like a fable and was really just liver tissue in a dish. He had human liver cells from older donors, behaving the way old cells behave, and he bathed them in blood plasma taken from people in their twenties. The old cells started making proteins the way young cells make them.

Something in young blood was telling old tissue to act young, and Pickart spent the next several years taking human plasma apart to find out what it was.

It turned out to be very small. Three amino acids, glycine, histidine and lysine, folded around a single copper atom. Glycyl-histidyl-lysine, GHK for short, GHK-Cu once the copper is attached. That's the copper peptide sitting in half the serums on the shelf at Sephora, and it entered the world as the answer to a question about liver cells in a dish.

It's a flare, not a fertilizer

Here's the part that changed how I read the marketing.

GHK isn't something your body manufactures on purpose and releases because it's feeling generous. It's a fragment. The sequence sits buried inside collagen and inside a protein called SPARC, and it only gets cut loose when that tissue is broken down, which is to say when you're hurt. Enzymes chew through damaged collagen, GHK falls out, it grabs whatever copper is nearby, and the molecule that results is read by every cell around it as an instruction. Something tore here. Start rebuilding.

Researchers call this kind of molecule a matrikine, a signal released by the structural matrix itself at the moment that matrix is damaged. Your body doesn't broadcast it. Your body fires it.

Which means every copper peptide serum on the market is, quite literally, selling you a distress signal in a bottle. Deliberately. The whole pitch, stripped of the packaging, is that if you can convince skin it has been injured, it will do what injured skin does, and what injured skin does is build.

And you fire fewer of them now

Pickart's group reported plasma GHK running around 200 nanograms per milliliter in a person's twenties and falling to roughly 80 by sixty. Those are old measurements that have been repeated for fifty years without much fresh work behind them, so hold the exact numbers loosely. The shape is what's worth having, and the shape matches what every woman past forty-five already knows without being told, which is that a scrape on the back of your hand now takes two weeks to do what it used to do in four.

It also lands in exactly the window where estrogen is dropping and skin collagen is going down with it. Two curves falling at once, in the same decade, for unrelated reasons. That's the honest reason this compound gets so much attention from women our age and so little from the twenty-five-year-olds who dominate the peptide forums.

The number that gets oversold

You'll see a claim repeated everywhere that GHK "resets 4,000 genes." It comes from real work, and it doesn't mean what the ad copy wants it to mean.

Pickart and Anna Margolina ran GHK's gene expression fingerprint through the Broad Institute's Connectivity Map, a reference library that compares what a compound does to cells against thousands of other compounds. They reported GHK shifting expression in 4,048 human genes, about six percent of the genome, and published it in 2014. The number is real.

What it describes is a signature in cultured cells. It isn't a woman putting serum on her face at night and moving four thousand genes in her skin, and nobody has shown that it is. The human skin evidence that does exist is thinner and much less thrilling, mostly small cosmetic-industry studies on topical creams, some of them decades old. A molecule can be genuinely fascinating in a dish and still be modest on a face, and GHK is currently both.

One honest line, said once. GHK-Cu is sold as a research compound, and there are no large human trials establishing a safe dose or a long-term profile in people. You already knew that. You're an adult making a call about your own body, and that call is yours.

Since we do this for a living

The price on this one is genuinely odd, so it's worth thirty seconds. Pull up the comparison tool and GHK-Cu runs from 29 cents to $4.43 a milligram across the 37 vendors that sell it. Fifteen to one, same compound, and almost none of it is about quality.

Most of the spread is bottle size. A small vial carries the same filling and handling cost as a big one spread across a fraction of the milligrams, so the per-milligram number climbs fast at the small end. Buy small and part of what you're buying is glass. The rest is simply which vendor you picked, which is the entire reason the tool sorts by milligram instead of by sticker price.
